Abstract

ABSTRACTThe Habitat Characteristic of the Critically Endangered Turtle the Sulawesi ForestSulawesi Turtle Leucocephalon yuwonoi in Ganonggol and Bangkir Rivers in CentralSulawesi. The Sulawesi Forest Turtle (Leucocephalon ymonoi) is an endemic, criticallyendangered and listed in the appendix I1 of CITES. Captive breeding is needed toreintroduce the species into the original habitat. Captive breeding success includes acomprehensive knowledge on the biology and ecology of the species. The objective ofthis research is to investigate the habitat character of the turtle respectively vegetation ofthe riverbank, physical and chemical conditions ofthe water. Field surveys were conductedon the Moutong area from 1 l until 19 October 2004 and on the Bangkir area from 20until 26 October 2004. Vegetation data were obtained by developing four quadrate at theturtle habitat in each locality and were analyzed using Curtis method. The results shownthat the habitat of Leucocephalon yuwonoi is transparent streaming water, neutral, nottoo high dissolved oxygen, water height not more than 50 cm, substrat varies from bigstones, small stones, clay loam and sandy loam. The area was covered by vegetation thatcan provide food during all year round, such as Colocasia esculenta, Limnocharisflava,and Ipomoea aquatics.Key Word: Leucocephalon yuwonoi, habitat, vegetation, water conditions.

Abstract

ABSTRACTSumatran Serow Capricornis sumatraensis sumatraensis (Bechstein, 1799))Distribution at Sipurak-Kerinci Seblat National Park. Sumatran serow [Capricornissumatraensis sumatraensis (Bechstein, 1799)J is one of endangered species in KerinciSeblat National Park (KSNP). Few data are available about Sumatran serow in KSNP.The research was designed to analize the distribution of Sumatran serow in Sipurak.Fieldstudy was conducted from September 2005 to February 2006 in Sipurak, KSNP.The detection-non detection survey method was applied for observation. There were 44cells sampling, the number of cells determined by purposive sampling base on habitattypes and accessibility to reach the location of cell (easy or hard). The data were analyzedby Arcview 3.2 PC. The sumatran serow just indirectly detected. Sign of its existencemainly detected at ramp. Distribution of sumatran serow mainly was invented on the lowland forest type (8 1,82%) which are characterized by caves, rock-cliff, and rugged hills.Key words: Distribution; detection-non detection; sumatran serow; Sipurak

Abstract

ABSTRAKParameter Kinetika Endokitinase dari Bacillus licheniformis MB-2. Bacilluslicheniformis MB-2 yang diisolasi dari air panas di Tompaso (Sulawesi Utara)menghasilkan 5 enzim kitinase ekstraselular. Salah satu enzim, Chi-67, telah dimumikandan diarekterisasi. Chi-67 dapat menghidrolisis substrat kitin buatan maupun alami.Analisis kinetika aktivitas enzim terhadap berbagai substrat kitin telah dilakukan. Enzimdapat memotong 4-methylumbelliferyl N, N-diacetylchitobioside {MUF(GINAc),}, 4-methylumbellifelyl N:N N-triacetylchitotrioside {MUF(GINAc),), glikol kitin, dankoloidal kitin, tetapi tidak dapat melepaskan senyawa MUF dari 4-methylumbelliferylN-acetyl-R-D-glucosamine {MUF(GINAc)}. Ini menunjukkan Chi-67 memiliki tipepernotongan endo. K u ~kaec epatan reaksi substrat menunjukkan pola profil Michaelis-Menten. Kecepatan reaksi maksimum (V,,) terhadap MUF(GlNAc), identik denganVmatxe rhadap MUF(GINAC)~ya itu 0.02 pM.mitil . VmaeXn zim terhadap koloidal kitinadalah 0.01 pM.h- atau lebih rendah 3 kali dari nilai VmaXpa da substrat glikol kitin (0.03M h ) Nilai Kcu, untuk koloidal kitin sebesar 8.01 h- atau hampir 2 kali lebih tinggidari glikol kitin (4.32 h-I) sementara Kc, untuk MUF(GlNAc),adalah 3.65 min- atau1,5 kali lebih tinggi dari Kc%,M UF(GINAc), yang hanya 2.90 min- . Afinitas Chi-67terhadap koloidal kitin, glikol kitin, MUF(GINAc),, dan MUF(GINAc), yang ditunjukkanoleh nilai konstanta Michaelis (K,) masing-masing 3.08 rng.ml-I, 0.32 mg.ml-I, 0.26 mMdan 0.1 mM.Kata kunci: Bakteri, endokitinase, enzim, dan stabilitas panas

Abstract

ABSTRACTGenetic Variation of Bat in the Genus Aethalops (Chiroptera: Pteropodidae) fromIndonesia: Analysis of 12SrRNA Gene of Mitochondria1 DNA. We report a geneticvariation on 12SrRNA gene to investigate haplotype and nucleotide diversity, as well aspopulation structure of bats in the genus Aethalops. Samples consisted of 21 individualsfrom 7 populations of 5 islands (Sumatra, Jawa, Kalimantan, Bali and Lombok) coveringall islands distribution of this genus in Indonesia. Total DNA was extracted from livertissues. Fragment 12SrRNA gene region of the mitochondria1 DNA were amplified byPolymerase Chain Reaction usingprimers of L 1091 and H 1478. Nucleotide sequenceswere determined to investigate genetic variation of this genus. Results indicated thatfrom 294 bp nucleotide sequences, 33 polymorphic sites with 13 haplotypes were foundamong 7 populations.Key Words: Variation, Bat, Aethalops , Chiroptera, Indonesia, 12SrRNAmitochondrial.

Abstract

ABSTRACTDistribution of Nitrifying Bacteria in Floodplain Lake. Case Study in Lake LoaKang Fishery Reserve, East Kalimantan. Lake Loa Kang is a fishery reserve locatedin a floodplain system of River Mahakam. This floodplain lakes consists of severalhabitat types among the others are backswamp deposit lakes represented by Lake LoaKang and Lake Balikpapan. This two floodplain lakes exchange water with RiverMahakam and one of its tributary, the River Pela. Nitrification is one key ofmicrobiological process of nitrogen cycle in inland water. The nitrifying bacteria functionalgroup that mediated the process can be detected and identified by enrichment culture.This study was aimed to reveal feature of nitrification process in several floodplainhabitat types of Lake Loa Kang Fishery Reserve (LLFR) system with emphasize on thespatial distribution pattern of nitrifying bacteria. The results ofthe study will be utilizedas limnological database for development of conservation and restoration of Pesut habitat.Water samplings are conducted for three times in 2003, those were in dry season (June),transitional (August) and the rainy (Sept.). Results of the study showed that the numberof nitrifying bacteria is highest at connecting channel between floodplain lakes (L. BalikPapan) and River Mahakam and the lowest in small size ephemeral pond in LLFR (Lopak).Ex situ simulation of this biogeochemical cycle indicate a duration of 13 - 21 days tocomplete the nitrification process. The number of nitritation and nitratation bacteria inLake Loa Kang in the dry season is smaller compared to the number in rainy season. Theduration of nitrification process in LLFR suspected to be influenced by the number ofbacteria, organic material and season.Key words: Distribution,Floodplain, Lake Loa Kang Fishery Reserve, Nitrifying bacteria
